Product Specific Information

The OKT8 monoclonal antibody reacts with the human CD8a molecule, a 32–34 kDa cell surface receptor expressed as either a heterodimer (CD8αβ) or homodimer (CD8αα). CD8a is found on thymocytes, mature T cells, and NK cells, and binds to MHC class I molecules. Through its association with protein tyrosine kinase p56lck, CD8a plays a role in T-cell development and activation.

OKT8, RPA-T8, and HIT8a clones do not compete for binding to human peripheral blood leukocytes, indicating distinct epitopes.

Applications Reported: Suitable for flow cytometric analysis.
Applications Tested: Prediluted and validated for flow cytometry using normal human peripheral blood cells. Recommended usage is 5 µL (0.125 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Alexa Fluor 532 is excited by the green laser (532 nm) and emits at 561 nm. Not compatible with yellow-green laser (561 nm). Recommended filter: 560/14 band pass.

Target Information

Cluster of Differentiation 8 (CD8) is a type I transmembrane glycoprotein of the immunoglobulin receptor family. It functions as a co-receptor with the T cell receptor (TCR) for MHC class I molecules, essential for T cell differentiation and activation. CD8 is expressed on cytotoxic T cells, NK cells, monocytes, and dendritic cells, forming disulfide-linked dimers of CD8α and CD8β or homodimers of CD8α.

Upon MHC-I/peptide complex engagement, CD8 recruits Lck kinase, promoting lymphokine production, motility, and cytotoxic activation, critical for pathogen and tumor cell clearance. Differentiation of naive CD8+ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
