Product Specific Information

The M5/114.15.2 monoclonal antibody reacts with mouse major histocompatibility complex (MHC) class II molecules, recognizing both I-A and I-E subregion-encoded glycoproteins (I-A^b, I-A^d, I-A^q, I-E^d, I-E^k). It detects a polymorphic determinant on B cells, monocytes, macrophages, dendritic cells, and activated T lymphocytes from mice carrying haplotypes H-2^b, H-2^d, H-2^q, H-2^p, H-2^r, and H-2^u but not H-2^s or H-2^f.

The M5/114 mAb can inhibit I-A–restricted T-cell responses in H-2^b, H-2^d, H-2^q, H-2^u haplotypes but not in H-2^f, H-2^k, or H-2^s.

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on mouse splenocytes; may be used at ≤0.125 µg/test (100 µL final volume). Optimal titration is recommended.

eFluor 506 is excited by the violet laser (405 nm) and emits at 506 nm. Recommended detection with a 510/20 band pass filter.

Target Information

MHC class II molecules are transmembrane glycoproteins expressed on professional antigen-presenting cells such as macrophages, dendritic cells, and B cells. They bind exogenous antigens within endocytic vesicles and present them to T cells. The antigen-binding groove between the α and β chains is open-ended and accommodates peptides 15–24 amino acids long.

HLA and MHC antibodies are important tools in Immunopeptidomics, aiding in the identification and characterization of neoantigens using high-performance liquid chromatography and tandem mass spectrometry.
