Product Specific Information

The monoclonal antibody 20Raj1 recognizes the human Ki-67 protein. Two isoforms of Ki-67 (345 and 395 kDa) are expressed in dividing cells. Ki-67 is present during active phases of the cell cycle (G1, S, G2, M) and absent in resting cells (G0). During interphase, it localizes to the nucleus, redistributing to chromosomes during mitosis and associating with heterochromatin-bound proteins such as CBX3.

In tumor cell studies, Ki-67 expression serves as a marker for proliferating cell fractions. This antibody also recognizes canine Ki-67.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-titrated for intracellular staining and flow cytometric analysis of stimulated normal human peripheral blood cells using the Foxp3/Transcription Factor Staining Buffer Set (Product #00-5523-00) and protocol.
Use at 5 µL (0.03 µg) per test (100 µL final volume). Suitable for 10⁵–10⁸ cells/test.

Fluorochrome Information:
eFluor® 506 is excited with the violet laser (405 nm) and emits at 506 nm. Recommended filter: 510/20 band pass or equivalent. Ensure instrument compatibility.

Filtration: 0.2 µm post-manufacturing filtered.

Target Information

Ki-67 is a nuclear protein expressed during late G1, S, G2, and M phases of the cell cycle. It contains a forkhead associated (FHA) domain and associates with euchromatin at the perichromosomal layer, centromeric heterochromatin, and nucleolus. Ki-67 shows cell cycle-dependent localization: perinucleolar in G1, nuclear matrix in G2, and chromosomal during M phase.

Ki-67 is a well-established proliferation marker, absent in G0 and increasing through mitosis. It is widely used to determine cell growth fractions in neoplasms, with prognostic value correlating with tumor grade. Also used as a neuronal marker for cell cycling and proliferation.
