Product Specific Information

The FJK-16s antibody reacts with mouse, rat, dog, porcine, bovine, and cat Foxp3 (FORKHEAD BOX P3, SCURFIN, JM2). Foxp3 is a 49–55 kDa transcriptional regulator involved in Treg cell function.
Immunoblotting with FJK-16s mapped the epitope to amino acids 75–125 of mouse Foxp3. In humans, this region is alternatively spliced.
Applications reported include intracellular staining followed by flow cytometric analysis. Tested using Foxp3/Transcription Factor Staining Buffer Set (Product # 00-5523-00) and BestProtocols® Protocol B. Recommended use ≤0.5 µg/test in 100 µL final volume.

Fluorochrome Information:
eFluor® 506 is excited by violet laser (405 nm) and emits at 506 nm. Recommended filter: 510/20 band pass or equivalent.
Filtration: 0.2 µm post-manufacturing filtered.

Target Information

FOXP3 (Forkhead box protein 3) is a key transcriptional regulator essential for immune homeostasis. It is highly expressed in CD25+CD4+ regulatory T cells, low in CD4+CD25− cells, and absent in CD8+ T cells.
FOXP3 mutations cause the scurfy phenotype in mice and IPEX syndrome in humans. Transcript variants encoding different isoforms have been identified. In human cancer cells, FOXP3 expression is regulated by p53 in response to DNA damage.
