Target Information
TAK1 (MAP3K7)는 serine/threonine kinase 계열에 속하며 다양한 신호전달 기능을 수행하는 단백질입니다.
TGF-β 활성화에 의해 처음 확인되었으며, TGF-β receptor 및 TRAF6과 상호작용하여 JNK와 p38의 활성화를 조절합니다.
또한 Wnt, BMP, activin, thyroid hormone, osmotic stress, endothelin, ephrine 등의 신호 경로에서도 p38 및 JNK를 활성화합니다.
TAK1은 von Hippel-Lindau tumor suppressor 발현을 통해 PDGF-B, integrin β1, β5를 억제하여 상처 치유를 촉진합니다.
IL-1, IL-6, IL-18, TNF, CD40, TLR, RIG-I 등 다양한 경로에서 IKK 활성화에 필수적인 역할을 하며, TAB2/3와의 결합 및 polyubiquitination을 통해 TAK1-IKK 경로를 활성화합니다.
TAK1은 또한 인간 telomerase reverse transcriptase를 억제하여 세포 수명 조절에도 관여합니다.
