Target Information

TAK1 (also known as MAP3K7) is a serine/threonine kinase involved in multiple signaling pathways. Initially identified as a TGF-β–activated kinase, TAK1 interacts with TGF-β receptor and TRAF6 to modulate activation of JNK and p38. It also participates in Wnt, BMP, and activin signaling, and responds to thyroid hormone, osmotic stress, endothelin, and ephrine. TAK1 promotes wound healing via von Hippel-Lindau tumor suppressor activation and regulates PDGF-B and integrin expression. It is essential in IKK activation through IL-1, IL-6, IL-18, TNF, CD40, TLR, and RIG-I pathways, requiring TAB2/3 and ubiquitination. TAK1 autophosphorylates at Thr178, Thr184, Thr187, and Ser192, subsequently phosphorylating IKK-β. Additionally, TAK1 represses human telomerase reverse transcriptase, suggesting a role in cell lifespan regulation.
