Product Specific Information
The WU24A4P monoclonal antibody reacts with human interleukin (IL)-17F homodimer (IL-17FF). This antibody specifically recognizes the IL-17FF homodimer and does not react with the IL-17AF heterodimer. IL-17FF is a 34 kDa cytokine belonging to the IL-17 family, which includes IL-17A, B, C, D, and E (IL-25). IL-17A and IL-17F are primarily produced by Th17 cells and play overlapping but distinct roles in immune regulation.
In vitro Th17 cells can express IL-17FF, IL-17AA, and IL-17AF dimers depending on culture conditions and differentiation state. These dimers signal through the IL-17RA/IL-17RC receptor complex and are implicated in inflammatory diseases such as inflammatory bowel disease, asthma, atopic dermatitis, and rheumatoid arthritis.
Clone WU24A4P is specific for IL-17FF homodimer, unlike other clones (e.g., SHLR17) that recognize IL-17F monomer in both IL-17FF and IL-17AF dimers.
Applications Reported: Suitable for intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-titrated and tested by intracellular staining and flow cytometry of Th17-polarized human peripheral blood cells using the Intracellular Fixation & Permeabilization Buffer Set (Product #88-8824-00) according to BestProtocols® Protocol A (Two-step protocol for cytoplasmic intracellular proteins).
Recommended usage: 5 µL (0.25 µg) per test. A test is defined as staining a cell sample in 100 µL final volume (typically 10⁵–10⁸ cells/test).
Fluorochrome Information:
eFluor® 660 replaces Alexa Fluor® 647. It emits at 659 nm and is excited with a red laser (633 nm). Ensure instrument compatibility.

Interleukin 17F (IL-17F) is a cytokine produced by inflammatory cells such as activated T cells, mast cells, and basophils. It contributes to allergic airway inflammation and induces cytokine, chemokine, and adhesion molecule expression in epithelial, endothelial, fibroblast, and eosinophil cells. IL-17F can form homodimers (IL-17FF) or heterodimers (IL-17AF) and signals via the IL-17R receptor, recruiting monocytes and neutrophils to inflammation sites.
