Description:
The 31SNEZE monoclonal antibody reacts with human IL-31, a member of the IL-6 cytokine family. IL-31 is expressed by CD4⁺ T cells (particularly Th2 phenotype) and mast cells. The IL-31 receptor is a heterodimer composed of IL-31RA and OSMRβ subunits, expressed by macrophages, monocytes, eosinophils, keratinocytes, and epithelial cells. IL-31 receptor signaling activates STAT3 and promotes release of proinflammatory cytokines and chemokines such as IL-1β, IL-6, IL-8, CXCL1, and CCL2. Overexpression of IL-31 is correlated with allergic asthma, rhinitis, and atopic dermatitis severity.

Applications Reported:
Validated for intracellular staining followed by flow cytometric analysis.

Applications Tested:
Pre-titrated for use in intracellular staining and flow cytometry of human peripheral blood lymphocytes. Recommended usage: 5 µL (0.125 µg) per 100 µL test volume. Cell number may range from 10⁵ to 10⁸ cells/test.

eFluor® 660 replaces Alexa Fluor® 647, emitting at 659 nm and excited by the red laser (633 nm). Ensure instrument compatibility.

Target Information

Interleukin-31 (IL-31) is a T-cell cytokine related to IL-6 family cytokines, preferentially produced by Th2 cells. IL-31 acts through a receptor complex of IL-31RA and oncostatin M receptor (OSMR). This interaction activates JAK/STAT and AKT signaling pathways. IL-31 receptor expression is constitutive in epithelial cells and inducible in activated monocytes. Overexpression in transgenic mice induces pruritus, alopecia, and skin lesions, implicating IL-31 in atopic dermatitis and as a potential antipruritic drug target.
