Product Specific Information

The HA58 monoclonal antibody reacts with human CD54 (InterCellular Adhesion Molecule-1, ICAM-1), a 90–110 kDa transmembrane glycoprotein expressed by monocytes, lymphocytes, and endothelial cells. CD54 expression increases on activated lymphocytes, and its interaction with ligand CD11a plays a key role in inflammatory responses.

Applications Reported:
Flow cytometry, Western blot, and immunohistochemistry. It has also been used to block in vitro mixed lymphocyte reactions (use Functional Grade HA58, Product #16-0549-82, for functional assays).

Applications Tested:
Tested in flow cytometry with normal human peripheral blood cells. Recommended ≤0.25 µg per test (final volume 100 µL). Optimal titration is recommended for specific assays.

Target Information

ICAM-1 (CD54) is an 85–110 kDa single-chain type I integral membrane glycoprotein composed of five immunoglobulin superfamily repeats, a transmembrane region, and a cytoplasmic domain. It binds to integrins CD11a/CD18 (LFA-1) and CD11b/CD18 (Mac-1), and serves as a receptor for Rhinovirus. ICAM-1 is expressed on activated endothelial cells, epithelial cells, fibroblasts, chondrocytes, lymphocytes, monocytes, macrophages, dendritic cells, and neutrophils. Expression increases during inflammation and is also observed in certain carcinoma and melanoma cells. Soluble ICAM-1 is detectable in plasma and elevated in inflammatory conditions.
