Product Specific Information
The J43 monoclonal antibody reacts with mouse PD-1 (programmed death-1), a 55 kDa member of the Ig superfamily. PD-1 contains the immunoreceptor tyrosine-based inhibitory motif (ITIM) and plays a key role in peripheral tolerance and autoimmune disease in mice. PD-1 is expressed mainly on activated T and B lymphocytes. PD-L1 (B7-H1) and PD-L2 (B7-DC) are identified as PD-1 ligands, showing overlapping functions and expression on normal tissues and activated antigen-presenting cells.
J43 inhibits the binding of mouse PD-L1-Ig and PD-L2-Ig to PD-1/BHK transfected cells. In vivo, J43 (intact or Fab) enhances contact hypersensitivity and exacerbates acute GVHD, EAE, and NOD diabetes, similar to PD-1-deficient cells or antibodies to PD-L1/PD-L2.

Applications Tested: Flow cytometric analysis of Con A-stimulated mouse splenocytes and PD-1 transfected cells.
Recommended usage ≤ 0.25 µg/test (100 µL final volume, 10⁵–10⁸ cells/test). Antibody titration recommended for optimal performance.
Target Information
Cell-mediated immune responses are initiated by T lymphocytes stimulated by peptides bound to MHC molecules on antigen-presenting cells (APC). Activated T cells express inhibitory receptors such as PD-1 (PDCD-1), which binds ligands PD-L1 and PD-L2 to suppress immune activation. PD-1 signaling prevents overactivation in the absence of danger signals (e.g., LPS). PD-1-deficient mice exhibit hyperactivated immune systems and autoimmune disorders. PD-1 often migrates at higher molecular weight in SDS-PAGE.
