Product Specific Information

The 12G5 monoclonal antibody reacts with human CD184 (CXCR4), also known as Fusin, LESTR, or HUMSTR. CD184 is a G-protein-coupled chemokine receptor with seven transmembrane domains and functions as a coreceptor for X4 HIV-1 entry into CD4+ cells.

CD184 is predominantly expressed on naive T cell subsets in peripheral blood and is rapidly upregulated by PHA and IL-2 stimulation. The 12G5 antibody partially inhibits SDF-1–induced chemotaxis and calcium influx, blocks CD4-independent HIV-2 infection, and inhibits CD4-dependent infection by certain T-tropic HIV-1 isolates.

Applications Tested: Tested on normal human peripheral blood cells at ≤0.5 µg/test (in 100 µL final volume). Optimal cell number: 10⁵–10⁸ cells/test. Titration recommended for optimal performance.

Target Information

CXCR4 (also known as Fusin, LESTR, HUMSTR) is a G-protein-coupled chemokine receptor involved in HIV-1 and HIV-2 entry into CD4+ cells. It binds to the chemokine CXCL12/SDF-1, mediating intracellular calcium increase and MAPK1/MAPK3 activation. CXCR4 also acts as a receptor for extracellular ubiquitin, influencing calcium and cAMP signaling.

CXCR4 is critical in hematopoiesis, cardiac septum formation, and vascular development in the gastrointestinal tract. In the CNS, it supports hippocampal neuron survival and cerebellar development. It mediates inflammatory responses to bacterial LPS, including TNF secretion by monocytes. Antibodies targeting CXCR4 can block HIV-1 and HIV-2 fusion and infection.
