Target Information
BCL-2 is a key regulator of apoptosis that can inhibit or promote cell death. Members of the BCL-2 family interact through dimerization to modulate apoptosis. Bag1 forms a heterodimer with BCL-2, enhancing its anti-apoptotic effect. Bax and Bak play critical roles in cytochrome c release from mitochondria, initiating apoptosis. Bax promotes apoptosis, whereas BCL-2 inhibits it. Constitutive expression of BCL-2, such as through translocation to the Ig heavy chain locus, is associated with follicular lymphoma. Neoplastic germinal centers in follicular lymphoma express high levels of BCL-2 alpha protein, unlike normal centers. Overexpression of BCL-2 has been linked to cancers such as B-cell lymphoma and prostate cancer.
