Target Information
ERN1 serine/threonine kinase (endoplasmic reticulum to nucleus signaling 1) is the ER to nucleus signaling 1 protein, a human homologue of the yeast Ire1 gene product. It possesses intrinsic kinase and endoribonuclease activities and plays a key role in endoplasmic reticulum-based stress signaling.
IRE1 acts as a sensor of unfolded proteins in the ER and initiates the unfolded protein response by splicing XBP1 mRNA, converting it into a potent transcriptional activator.
Null mutants are viable but IRE1 is essential under stress conditions that cause unfolded protein accumulation in the ER. Depletion of IRE1p through dominant negative expression leads to apoptosis under such stress conditions.
