Target Information
Tumor necrosis factor (TNF) receptor associated factors (TRAFs) are adaptor proteins linking the TNF receptor superfamily to signaling pathways, playing key roles in regulating cell death and stress responses. TRAF proteins share a conserved domain enabling receptor and TRAF interactions, activating various signaling cascades.
TRAF2 and TRAF3 bind directly to CD40, essential for NF-kappa-B activation in mouse B lymphocytes. TRAF2, together with TRAF6, is required for CD40 signaling in non-hematopoietic cells and interacts with the lymphotoxin-beta receptor (LTβR) along with TRAF3 and apoptosis inhibitors such as cIAP1 and Smac.
