Target Information

ERN1 (endoplasmic reticulum to nucleus signaling 1) encodes the ER to nucleus signaling protein, a human homologue of the yeast Ire1 gene product.
This protein possesses intrinsic kinase and endoribonuclease activity and plays a key role in ER-based stress signaling.
IRE1 acts as a sensor of unfolded proteins in the ER and transmits the unfolded protein signal to the nucleus by splicing XBP1 mRNA, converting it into a potent transcriptional activator.
It is essential for fetal viability and cell survival under stress conditions that cause unfolded protein accumulation in the ER.
Depletion of IRE1p through dominant negative expression leads to apoptosis under such stress conditions.
