Immunohistochemistry (PFA fixed): incubate antibody for 30 minutes at room temperature.
Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 minutes at 95°C followed by cooling at room temperature for 20 minutes.
Target Information
Apolipoprotein B (apo B) in human plasma is a major protein of low-density lipoproteins (LDL) with a molecular mass of approximately 260–500 kDa. Apolipoprotein B binds to specific receptors on cell membranes and is involved in removal of LDL and very low-density lipoprotein (VLDL) cholesterol from circulation.
LDL acts as the carrier protein for cholesterol in the blood, binding to its receptor on capillary walls and mediating uptake and clearance of cholesterol. In atherosclerotic lesions, oxidatively modified LDL is found and recognized by macrophages via scavenger receptors A and CD36. Oxidized LDL may serve as a marker of atherosclerosis.
Apolipoprotein B is primarily synthesized in the liver and is a major component of very low-density, intermediate-density, and low-density lipoproteins, as well as lipoprotein(a). It functions as a ligand for the LDL receptor, and elevated levels are associated with premature atherosclerosis. Normal plasma Apolipoprotein B levels are around 800 mg/L.
Apolipoprotein B occurs as two main isoforms, apoB-48 and apoB-100, encoded by a single gene. The shorter apoB-48 protein results from RNA editing of the apoB-100 transcript at residue 2180, creating a stop codon and terminating translation early. Mutations in the Apolipoprotein B gene or its regulatory region cause disorders such as hypobetalipoproteinemia and hypercholesterolemia due to ligand-defective Apolipoprotein B.
