Product Specific Information

  • Immunohistochemistry (PFA fixed):
    Incubate antibody for 30 minutes at room temperature.
    Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

Apolipoprotein B (apo B) in human plasma is a major protein of low-density lipoproteins (LDL) with a molecular mass of approximately 260–500 kDa.
Apolipoprotein B binds to specific receptors on cell membranes and is involved in the removal of LDL and very low-density lipoprotein (VLDL) cholesterol from circulation.

Low-density lipoprotein (LDL) is the carrier protein for cholesterol in the blood. LDL binds to its receptor on capillary walls, mediating cholesterol uptake and clearance.
In atherosclerotic lesions, oxidatively modified LDL is found and recognized by macrophages via scavenger receptor A and CD36.
Oxidized LDL may serve as a marker of atherosclerosis. Apolipoprotein B is synthesized mainly in the liver and is a major component of VLDL, IDL, LDL, and lipoprotein(a).
It acts as a ligand for the LDL receptor, and elevated levels are associated with premature atherosclerosis.
Normal plasma apolipoprotein B levels are around 800 mg/L.

Apolipoprotein B occurs in plasma as two main isoforms: apoB-48 and apoB-100.
Both are encoded by a single gene, with apoB-48 produced by RNA editing of the apoB-100 transcript, introducing a stop codon at residue 2180.
Mutations in the Apolipoprotein B gene or regulatory regions can cause hypobetalipoproteinemia and hypercholesterolemia due to ligand-defective Apolipoprotein B.
