Product Specific Information

Description:
The eBioSN4 monoclonal antibody reacts with human CD9, a 24 kDa tetraspanin family protein characterized by four hydrophobic transmembrane domains. CD9 is expressed in platelets, eosinophils, basophils, pre-B cells, activated T cells, and neural cell lines. Its expression is associated with malignant phenotypes, including 90% of non-T cell acute lymphoblastic leukemia and 50% of chronic lymphocytic and acute myeloblastic leukemias.
In platelets, CD9 is localized in alpha-granules and interacts with integrin αIIb/βIII, contributing to platelet aggregation. CD9 also promotes aggregation of pre-B cell lines and mediates adhesion and migration of pre-B and Schwann cells. It can co-stimulate T cells independently of CD28. The eBioSN4 antibody partially cross-blocks binding of the anti-human CD9 antibody MM2/57.

Applications Reported:
Flow cytometric analysis.

Applications Tested:
Pre-titrated and validated for flow cytometry using normal human peripheral blood cells. Recommended usage: 5 µL (0.125 µg) per 100 µL test volume. Cell count range: 10⁵–10⁸ cells/test.

Target Information

CD9 antigen is a surface glycoprotein expressed on developing B lymphocytes, platelets, monocytes, eosinophils, basophils, stimulated T lymphocytes, neurons, and glial cells. It belongs to the tetraspanin family, which spans the membrane four times.
In pre-B cells and platelets, CD9 regulates activation and aggregation via association with integrin CD41/CD61 (GPIIb/GPIIIa). It plays roles in cell motility, osteoclastogenesis, neurite outgrowth, myotube formation, and sperm-egg fusion. CD9 is essential for MHC II molecule association on dendritic cells, influencing T cell stimulation.
CD9 functions in differentiation, adhesion, and signal transduction, contributing to suppression of cancer cell motility and metastasis, and is considered a metastasis suppressor in solid tumors.
