Description:
The RPA-T8 monoclonal antibody reacts with the human CD8a molecule (32–34 kDa), a cell surface receptor expressed as a heterodimer with CD8β or as a homodimer (CD8α/α). It is present on most thymocytes and a subset of mature T cells and NK cells. CD8 binds MHC class I and associates with tyrosine kinase p56lck, playing a key role in T-cell development and activation.

Reported for use in flow cytometric analysis.
Applications Tested:
Pre-titrated and tested for flow cytometric analysis of normal human peripheral blood cells. Use at 5 µL (1 µg) per test in a final volume of 100 µL. Cell numbers may vary from 10⁵ to 10⁸ cells/test.

Target Information
Cluster of differentiation 8 (CD8) is a type I transmembrane glycoprotein of the immunoglobulin receptor family. It functions as a co-receptor with the T-cell receptor (TCR) for MHC class I molecules. CD8 is expressed mainly on cytotoxic T lymphocytes (CTLs), as well as subsets of NK cells, monocytes, and dendritic cells. Upon binding to MHC-I/peptide complexes, CD8 recruits Lck kinase, leading to lymphokine production, motility, and CTL activation. Activated CTLs are key in pathogen and tumor cell clearance. Differentiation of naive CD8⁺ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
