Product Specific Information

The OKT8 monoclonal antibody reacts with the human CD8a molecule, a 32–34 kDa cell surface receptor expressed as a heterodimer (CD8αβ) or homodimer (CD8αα). CD8a is found on most thymocytes and a subset of mature T cells and NK cells. It binds to MHC class I and associates with protein tyrosine kinase p56lck, playing a key role in T-cell development and activation.

Preliminary testing shows that OKT8 does not compete with other anti-human CD8 clones (RPA-T8, HIT8a) for binding, suggesting distinct epitopes.

Applications Tested: Pre-titrated for flow cytometry of normal human peripheral blood cells. Use 5 µL (0.06 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Target Information

CD8 (Cluster of Differentiation 8) is a type I transmembrane glycoprotein of the immunoglobulin receptor family. It functions as a co-receptor for MHC class I molecules and is crucial for T cell differentiation and activation. CD8 is mainly expressed on cytotoxic T lymphocytes and a subset of NK cells, monocytes, and dendritic cells. Upon MHC-I/peptide complex binding, CD8 recruits Lck kinase, initiating signaling that leads to cytokine production, motility, and cytotoxic T lymphocyte activation. Differentiation of naïve CD8⁺ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
