Product Specific Information

This antibody recognizes phosphorylated intracellular tyrosine 142 of CD3 zeta chain (CD247), a component of the TCR/CD3 complex expressed on T cells.

• Flow cytometry: Intracellular staining
• Western blotting: Non-reducing conditions

Target Information

The CD3z complex is essential for transducing antigen-recognition signals into the cytoplasm of T cells and regulating surface expression of the TCR complex. T cell activation through the antigen receptor involves CD3 subunits (gamma, delta, epsilon, zeta) — structurally related members of the immunoglobulin superfamily encoded on human chromosome 11.
CD3 subunits have long cytoplasmic tails that interact with signal transduction molecules via double tyrosine-based motifs. CD3 is involved in TCR-induced growth arrest, cell survival, and proliferation.

CD3 antigen is expressed on:

• 68–82% of normal peripheral blood lymphocytes
• 65–85% of thymocytes
• Purkinje cells in the cerebellum

It is not expressed on B or NK cells. Decreased T lymphocyte percentages may be observed in autoimmune diseases.
Diseases associated with CD3z include Immunodeficiency 25 and T-B+ Severe Combined Immunodeficiency due to CD3δ/ε/ζ defects.
