• For IHC (PFA-fixed): incubate antibody for 30 minutes at room temperature.
• Staining of formalin-fixed tissues requires heating sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

CD30 (Ki-1, TNF Receptor Superfamily Member 8) is a type I transmembrane glycoprotein of the TNF receptor superfamily. It was originally identified as a surface antigen of Hodgkin’s and Reed-Sternberg cells using monoclonal antibody Ki-1. The ligand for CD30 is CD30L (CD153), and their interaction mediates cell proliferation, activation, differentiation, and apoptosis.

CD30 plays a critical role in the pathophysiology of Hodgkin’s disease and other CD30+ lymphomas. It acts as a costimulatory molecule in thymic negative selection and is expressed on Hodgkin’s and Reed-Sternberg cells, some non-Hodgkin’s lymphomas, virus-infected T and B cells, and activated normal T and B cells.

In T cells, CD30 is expressed on subsets producing Th2-type cytokines and on CD4+/CD8+ thymocytes co-expressing CD45RO and IL4 receptor. Soluble CD30 (sCD30) reflects Th2 immune response. TRAF2 and TRAF5 interact with CD30 to activate NF-κB signaling. CD30 positively regulates apoptosis and limits autoreactive CD8 effector T cell proliferation, protecting against autoimmunity. Two alternatively spliced transcript variants encode distinct isoforms. CD30 is expressed by mononuclear cells in Hodgkin’s lymphoma, Reed-Sternberg cells, most Anaplastic Large Cell Lymphomas (ALCL), and embryonal carcinomas.
