Target Information
HIF1-alpha (HIF1A) is a subunit of HIF1, a transcription factor found in mammalian cells under low oxygen tension. HIF-1 is a heterodimer consisting of alpha and beta subunits, both belonging to the PAS family of transcription factors. It regulates the adaptive response to hypoxia by activating transcription of genes involved in oxygen delivery and metabolic adaptation, such as erythropoietin, glucose transporters, glycolytic enzymes, and VEGF.
HIF1-alpha also plays a role in hypoxia-mediated apoptosis, cell proliferation, and tumor angiogenesis. Under hypoxic conditions, HIF1-alpha interacts with p53 and undergoes MDM2-mediated ubiquitination and proteasomal degradation. Induction of NOX4 by HIF1-alpha helps maintain ROS levels after hypoxia. In humans, HIF1A is located on chromosome 14q, and its C-terminal region binds to p300, forming complexes that induce hypoxia-responsive genes.
Hypoxia contributes significantly to diseases such as myocardial and cerebral ischemia, cancer, pulmonary hypertension, congenital heart disease, and COPD.
