Product Specific Information

The HIP8 monoclonal antibody reacts with the human CD41 molecule (integrin αIIb, platelet GPIIb). CD41 associates with integrin β3 (CD61) and is expressed by megakaryocytes and platelets. The CD41/CD61 complex acts as a receptor for fibronectin, fibrinogen, von Willebrand factor, vitronectin, and thrombospondin, mediating platelet aggregation. HIP8 blocks platelet aggregation.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells (5 µL/test). A test is defined as the amount of antibody that stains a cell sample in 100 µL final volume. Cell number may range from 10⁵ to 10⁸ cells/test.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining with NovaFluor conjugates.
• Add 5 µL per sample before combining with cells. For single-color controls, use 5 µL per 100 µL cell sample.

NovaFluor conjugates utilize Phiton technology with engineered fluorescent signatures minimizing spillover and spread impacts.

Target Information

CD41 (platelet glycoprotein IIb, ITGA2B) consists of two subunits (α: 120 kDa, β: 23 kDa) that interact with CD61 in the presence of calcium to form a functional adhesive receptor. CD41 plays a role in platelet aggregation and blood coagulation by binding to von Willebrand factor, fibrinogen, fibronectin, and vitronectin. Expressed mainly on megakaryocyte-platelet lineage, CD41 is also present during early hematopoietic differentiation.
Diseases associated with CD41 dysfunction include Glanzmann Thrombasthenia and Platelet type-16 bleeding disorder.
