Product Specific Information
The HIP8 monoclonal antibody reacts with the human CD41 molecule (integrin alphaIIb, platelet GPIIb). CD41 associates with integrin beta3 (GPIIIa, CD61) and is expressed by megakaryocytes and platelets. The CD41/CD61 complex acts as a receptor for fibronectin, fibrinogen, von Willebrand factor, vitronectin, and thrombospondin, mediating platelet aggregation. HIP8 blocks platelet aggregation.
Each product includes:
- 1 vial of NovaFluor conjugate
- 1 vial of CellBlox Plus Blocking Buffer
Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells.
Use 5 µL (0.6 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).
Important Notes:
- NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
- Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.
- Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
- Add 5 µL per sample to antibody cocktails before mixing with cells.
- For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

Target Information
CD41 (platelet glycoprotein IIb, ITGA2B) consists of two subunits (120 kDa alpha and 23 kDa beta) that interact with CD61 in the presence of calcium to form a functional receptor. CD41 mediates platelet aggregation and binds von Willebrand factor, fibrinogen, fibronectin, and vitronectin upon vascular injury. It is primarily expressed on megakaryocyte-platelet lineage and is involved in early hematopoietic differentiation.
Associated diseases include Glanzmann Thrombasthenia and Platelet type-16 bleeding disorder.
