Product Specific Information
The HIP8 monoclonal antibody reacts with the human CD41 molecule (integrin αIIb, platelet GPIIb). CD41 associates with integrin β3 (CD61) and is expressed by megakaryocytes and platelets. The CD41/CD61 complex acts as a receptor for fibronectin, fibrinogen, von Willebrand factor, vitronectin, and thrombospondin, mediating platelet aggregation. HIP8 blocks platelet aggregation.
Each product contains:
- 1 vial of NovaFluor conjugate
- 1 vial of CellBlox Plus Blocking Buffer
Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells at 5 µL (0.6 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).
Important Notes:
- NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
- Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.
- Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining with NovaFluor conjugates.
- Add 5 µL per sample to antibody cocktails before combining with cells.
NovaFluor Technology:
Based on Phiton™ technology utilizing nucleic acid dye structures for engineered fluorescent signatures with minimized spillover and spread impacts.
Excitation: 552 nm; Emission: 663 nm; Laser: 561 nm (Yellow).
Target Information
CD41 (platelet glycoprotein IIb, ITGA2B) consists of α (120 kDa) and β (23 kDa) subunits that interact with CD61 in the presence of calcium to form an adhesive receptor. CD41 mediates platelet aggregation and binds von Willebrand factor, fibrinogen, fibronectin, and vitronectin upon vessel damage. Expressed on megakaryocyte-platelet lineage, CD41 is also found during early hematopoietic differentiation. Dysfunction is linked to Glansmann Thrombasthenia and Platelet type-16 bleeding disorder.
