Product Specific Information

This TNKUPJ monoclonal antibody recognizes mouse CD163, a 130 kDa surface receptor expressed by subsets of tissue macrophages such as splenic red pulp macrophages, Kupffer cells, and intestinal lamina propria macrophages. Mouse monocytes do not express CD163, and its expression pattern differs from human CD163, which is inducible by M2 cytokines.

This antibody can detect CD163 in fixed and permeabilized cells, enabling intracellular staining. For samples subjected to aggressive tissue dissociation, intracellular detection is recommended.

Applications Tested: Tested on mouse splenocytes at ≤0.125 µg/test (100 µL final volume per test). Optimize titration for best performance.

Fluorochrome Characteristics:
PE-eFluor 610 can be excited by 488–561 nm lasers and emits at 607 nm. Recommended detection filter: 610/20 band pass (PE-Texas Red equivalent). Protect from light due to photo-induced oxidation.

Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-57) for up to 3 days at 4°C in the dark with minimal signal loss.

Target Information

CD163 (M130 antigen, Ber-Mac3, Ki-M8, SM4) is a 130 kDa membrane glycoprotein, part of the scavenger receptor cysteine-rich superfamily, and serves as a receptor for hemoglobin-haptoglobin complexes. It protects tissues from oxidative damage by mediating uptake and recycling of iron.

CD163 is expressed on human monocytes and macrophages, particularly in late inflammation phases. Expression is upregulated by IL-10, IL-6, and dexamethasone, while LPS and PMA induce shedding from the cell surface. CD163 binding is calcium- and pH-dependent and triggers intracellular signaling cascades involving calcium mobilization and cytokine secretion (IL-6, CSF1).
