Beta-catenin is an adherens junction (AJ) protein identified as a component of cell-cell adhesion structures. AJs are essential for the formation and maintenance of epithelial cell layers by regulating cell growth and adhesion between cells. Beta-catenin interacts with the cytoplasmic domain of E-cadherin, linking it to alpha-catenin, which anchors the E-cadherin complex to the cortical actin cytoskeleton.

Beta-catenin also binds to cytoskeletal complexes containing the adenomatous polyposis coli protein and microtubules, and participates in signaling pathways involving tyrosine kinases, phosphatases, and Wnt/Wingless. These interactions regulate morphogenesis and cell proliferation.

It is expressed in various hair follicle cell types and epithelial tissues. Pathological roles have been identified in pilomatrixoma, medulloblastoma, colorectal cancer, ovarian cancer, and other solid tumors. In the nucleus, beta-catenin co-activates Lef/Tcf transcription factors, promoting expression of genes such as cyclin D and c-myc that drive cell proliferation.
