Product Specific Information

Heat-mediated antigen retrieval is recommended prior to staining, using a 10 mM citrate buffer (pH 6.0) for 10 minutes followed by cooling at room temperature for 20 minutes.
Following antigen retrieval, incubate samples with primary antibody for 30 minutes at room temperature.
A suggested positive control is anaplastic lymphoma.


Target Information

Anaplastic Lymphoma Receptor Tyrosine Kinase (ALK) belongs to the insulin receptor superfamily and is vital for brain development.
Mutations, rearrangements, and amplifications in the ALK gene have been identified in tumors such as anaplastic large cell lymphoma, neuroblastoma, and non-small cell lung cancer.
The most common genetic alteration is chromosomal rearrangement, creating a fusion gene between ALK (chromosome 2) and NPM (chromosome 5), producing the oncogenic NPM-ALK fusion protein.
ALK encodes a receptor protein-tyrosine kinase with transmembrane and extracellular domains, which are absent in the NPM-ALK fusion product.
ALK shows high sequence similarity to LTK and plays a key role in neuronal development and signaling.
