Product Specific Information

Heat-mediated antigen retrieval is recommended prior to staining using a 10 mM citrate buffer (pH 6.0) for 10 minutes, followed by cooling at room temperature for 20 minutes.
After antigen retrieval, incubate samples with the primary antibody for 10 minutes at room temperature.
A suggested positive control is hepatocellular carcinoma or fetal liver.

Target Information

AFP (Alpha Fetoprotein) is a major plasma protein produced by the yolk sac, intestinal tract, and liver during fetal life.
In adults, AFP expression is often associated with hepatoma or teratoma, though hereditary persistence may occur without pathology.
AFP is considered the fetal counterpart of serum albumin, with both genes located tandemly on chromosome 4.
AFP exists in monomeric, dimeric, and trimeric forms and can bind copper, nickel, fatty acids, and bilirubin.
AFP levels in amniotic fluid are used to screen for spina bifida and anencephaly.
Elevated serum AFP is associated with hepatocellular carcinoma, teratoblastoma, colorectal cancer, pancreatic cancer, and germ cell neoplasms.
