Product Specific Information
Heat-mediated antigen retrieval is recommended prior to staining, using a 10 mM citrate buffer (pH 6.0) for 10–20 minutes, followed by cooling at room temperature for 20 minutes.
After antigen retrieval, incubate samples with primary antibody for 30 minutes at room temperature.
A suggested positive control is normal breast tissue.
Target Information
BAX is a member of the Bcl-2 family and plays an important role in the regulation of apoptosis.
Whereas Bcl-2 is commonly regarded as an anti-apoptotic protein, BAX has a pro-apoptotic function.
Regulation of apoptosis involves both homo- and heterodimerization of different isoforms of BAX and Bcl-2.
The Bax gene encodes isoforms Bax alpha (21 kDa) and Bax beta (24 kDa). Both contain BH1, BH2, and BH3 domains; Bax beta has a unique carboxyl terminus lacking a hydrophobic transmembrane domain.
Bcl-2 also exists in multiple isoforms. Bcl-2 beta differs in the 3′ UTR and coding region compared to variant alpha, being shorter (22 kDa) and having a distinct C-terminus compared to Bcl-2 alpha (26 kDa).
Overexpression of BAX promotes cell death. BAX can form homodimers and heterodimerize with other BCL-2 related proteins.
