Target Information

Programmed death-ligand 2 (PD-L2, also known as B7-DC) is a member of the B7 ligand family within the immunoglobulin superfamily. Along with PD-L1, it acts as a ligand for PD-1, an inhibitory immune checkpoint molecule.
PD-L2 is primarily expressed in dendritic cells but can be induced in various immune and non-immune cells depending on the microenvironment. Its expression increases under Th2 cytokine IL-4 and, to a lesser extent, Th1 cytokines TNF-α and IFN-γ.
Although PD-L2 is generally expressed at lower levels than PD-L1, it has 2–6 times higher relative affinity to PD-1. Binding of PD-L2 or PD-L1 to PD-1 inhibits T-cell activation, proliferation, and cytokine secretion (e.g., IFN-γ, IL-10), providing negative feedback in immune regulation.
Recent studies indicate PD-L2 also interacts with RGMb (Repulsive Guidance Molecule b), influencing respiratory tolerance development in the lung through BMP and/or neogenin signaling pathways.
The human PD-L2 monomer consists of a 201-amino-acid extracellular domain, a 21-amino-acid transmembrane domain, and a 32-amino-acid cytoplasmic domain.
