Product Specific Information

The monoclonal antibody 20Raj1 recognizes the human Ki-67 protein. Two isoforms (345 and 395 kDa) are expressed in dividing cells, detectable during active phases (G1, S, G2, M) but absent in resting cells (G0).
During interphase, Ki-67 localizes to the nucleus and redistributes to chromosomes during mitosis, associating with heterochromatin-bound proteins such as CBX3.
Ki-67 expression is widely used as a marker for determining the fraction of proliferating cells in tumor studies.

This antibody also recognizes canine Ki-67.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-diluted and tested using the Foxp3/Transcription Factor Staining Buffer Set (Product # 00-5523-00).
Use at 5 µL (0.25 µg) per test (100 µL final volume). Cell number range: 10⁵–10⁸ cells/test.

Light Sensitivity: Protect vial and stained samples from light.
Fixation: Samples can be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-57).
Excitation: 488–561 nm; Emission: 695 nm; Laser: Blue, Green, Yellow-Green.

Target Information

Ki-67 is a nuclear protein expressed during late G1, S, G2, and M phases of the cell cycle. It contains a forkhead-associated (FHA) domain and associates with euchromatin and heterochromatin regions.
Expression pattern changes throughout the cell cycle:

• G1: Perinucleolar localization
• G2: Nuclear matrix localization
• M phase: Chromosomal association

Ki-67 serves as a proliferation marker since it is absent in G0 cells and increases from G1 through mitosis.
Used to determine the growth fraction in neoplasms and as a prognostic marker correlating with tumor grade.
Also applied as a neuronal marker for cell cycling and proliferation.
