Product Specific Information

The eBioC1.7 monoclonal antibody reacts with human CD244 (2B4, p38). CD244 is a 38 kDa protein expressed on NK cells, CD8+ T cells, γδ T cells, monocytes, basophils, and eosinophils. Binding of CD244 ligand CD48 results in NK cell activation and increased cytotoxicity and IFN-γ secretion.

Binding of C1.7 to CD244 induces tyrosine phosphorylation and recruitment of SLAM-associated protein (SAP). Patients with X-linked lymphoproliferative disorder (XLPD) have a mutation in SAP that prevents binding to phosphorylated CD244.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested by flow cytometry using human peripheral blood cells. Recommended use: 5 µL (0.25 µg) per test (100 µL final volume). Cell number range: 10⁵–10⁸ cells/test.

Fixation: Samples can be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-57) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.

Target Information

CD244 (2B4, SLAMF4) is a cell surface receptor expressed on NK cells and some T cells. It mediates non-MHC restricted killing and interacts with SLAMF2 to activate both SLAMF4- and SLAMF2-expressing cells. Lower SLAMF4 levels have been observed in systemic lupus erythematosus, suggesting a role in autoimmune pathology. Diseases associated with CD244 dysfunction include rheumatoid arthritis and lymphoproliferative syndrome. Multiple alternatively spliced transcript variants exist.
