Target Information
Pantothenate kinase (PANK2)는 CoA 생합성에서 중요한 조절 효소로, pantothenate (비타민 B5), N-pantothenoylcysteine, pantetheine의 세포질 인산화를 촉매합니다. CoA는 주요 아실 운반체로서 중간대사 및 지방산 대사에서 중심적인 역할을 합니다.
효모와 초파리에서는 pantothenate kinase 유전자가 하나뿐이며, 이 유전자가 결손되면 생존이 불가능합니다.
PANK2 유전자 돌연변이는 pantothenate kinase-associated neurodegeneration (PKAN, 이전 명칭 Hallervorden-Spatz syndrome, HSS)의 원인으로 알려져 있습니다.
PKAN은 뇌 내 철 축적과 관련된 상염색체 열성 신경퇴행성 질환입니다.
또한 PANK2 돌연변이는 hypoprebetalipoproteinemia, acanthocytosis, retinitis pigmentosa 및 pallidal degeneration (HARP)의 원인이 되기도 합니다.
