Product Specific Information

The OKT8 monoclonal antibody reacts with the human CD8a molecule, a 32–34 kDa cell surface receptor expressed as CD8αβ heterodimer or CD8αα homodimer. It is found on thymocytes, mature T cells, and NK cells. CD8 binds MHC class I and associates with protein tyrosine kinase p56lck, playing a key role in T-cell development and activation.

This antibody has been pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. Recommended use: 5 µL (0.2 µg)/test in 100 µL final volume.

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.
• Includes 1 vial of NovaFluor conjugate and 1 vial of CellBlox Plus Blocking Buffer (Product # C001T06F01).
• Add 5 µL of CellBlox Plus Blocking Buffer per sample for optimal staining.

Target Information

CD8 is a type I transmembrane glycoprotein of the immunoglobulin family functioning as a co-receptor for MHC class I molecules. It is expressed on T cells, NK cells, monocytes, and dendritic cells. Upon MHC-I/peptide binding, CD8 recruits Lck kinase, leading to activation of cytotoxic T lymphocytes (CTLs). CD8+ T cell differentiation is enhanced by IL-2, IL-12, and TGF-β1.
