Product Specific Information

The OKT8 monoclonal antibody reacts with the human CD8a molecule (~32–34 kDa), expressed as heterodimer (CD8αβ) or homodimer (CD8αα). It is found on thymocytes, subsets of mature T cells, and NK cells. CD8 binds to MHC class I and associates with tyrosine kinase p56lck, playing roles in T-cell development and activation.

Preliminary testing shows OKT8 does not compete with clones RPA-T8 or HIT8a for binding to human leukocytes, indicating distinct epitopes.

Each product includes:

• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated for flow cytometry on human peripheral blood cells (5 µL/test). A test corresponds to 100 µL final volume, with 10⁵–10⁸ cells/test.

Important Notes:

• NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability.
• CellBlox Plus Blocking Buffer (Product # C001T06F01) is required for optimal staining.
  • Add 5 µL per sample before mixing with cells.
  • For single-color controls: 5 µL per 100 µL cell sample (10³–10⁸ cells).

Target Information

CD8 (Cluster of Differentiation 8) is a type I transmembrane glycoprotein functioning as a co-receptor with TCR for MHC class I molecules. It is mainly expressed on cytotoxic T cells as a CD8αβ heterodimer, and on subsets of T cells, NK cells, monocytes, and dendritic cells as CD8αα homodimers.

Upon MHC-I/peptide binding, CD8 recruits tyrosine kinase Lck, triggering cytokine production, motility, and cytotoxic T lymphocyte (CTL) activation. Activated CTLs are essential for pathogen and tumor clearance. Differentiation of naïve CD8⁺ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
