Target Information

Zhou et al. have identified a protein that inhibits multinucleate osteoclast formation in murine osteoblast and spleen cell co-cultures. This protein, Osteoclast Inhibitory Lectin (OCIL), also inhibits osteoclast formation in spleen cell cultures treated with RANKL and M-CSF.
mOCIL acts directly on macrophage/monocyte cells, showing inhibitory action on adherent spleen cell cultures depleted of stromal and lymphocytic cells. It completely inhibited osteoclast formation during the proliferative phase and resulted in 70% inhibition during the differentiation phase.
OCIL mRNA expression in osteoblasts is enhanced by parathyroid hormone, calcitriol, interleukin-1, IL-11, and retinoic acid. OCIL is expressed in osteoblasts, chondrocytes, and various extraskeletal tissues. Since OCIL expression parallels RANKL distribution, it is suggested that RANKL and OCIL may interact in skeletal and extracellular tissues.
