Product Specific Information

Description:
The UCHT2 monoclonal antibody reacts with human CD5, a 67 kDa protein expressed by most thymocytes, mature T cells, and a subset of B cells. CD5 signaling activates T cells and mediates T-B cell interactions via CD72 binding. The UCHT2 antibody recognizes primate CD5.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported:
Flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of normal human peripheral blood cells. Recommended use ≤ 5 µL (0.4 µg) per 100 µL cell sample (10⁵–10⁸ cells/test). Optimize antibody titration for best performance.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
• Add 5 µL of CellBlox Plus per sample before combining with cells.
• For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology with engineered fluorescent signatures designed to minimize spillover and spread.
Excitation: 509 nm; Emission: 665 nm; Laser: 488 nm (Blue).

Target Information

CD5 is a 67 kDa human T-lymphocyte single-chain transmembrane glycoprotein expressed on all mature T cells, most thymocytes, and many T-cell leukemias and lymphomas. It is also found on a subset of activated B cells and may regulate T-cell proliferation. CD5 is expressed on approximately 95% of thymocytes and 72% of peripheral blood lymphocytes. In lymph nodes, expression is mainly in T cell areas.
Diseases associated with CD5 dysfunction include thymus cancer and Richter’s Syndrome.
