Product Specific Information

The UCHT2 monoclonal antibody reacts with human CD5, a 67 kDa protein expressed by most thymocytes, mature T cells, and a subset of B cells. CD5 signaling activates T cells and mediates T-B cell interactions via CD72 binding.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Suitable for flow cytometric analysis.
Applications Tested: Tested on normal human peripheral blood cells at ≤5 µL (0.4 µg) per test (100 µL final volume). Cell number range: 10⁵–10⁸ cells/test. Antibody titration recommended for optimal performance.

Important Notes:

• NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
  • Add 5 µL per sample before combining with cells.
  • For single-color controls: 5 µL per 100 µL of cell sample (10³–10⁸ cells).

Target Information

CD5 is a 67 kDa human T-lymphocyte transmembrane glycoprotein expressed on most thymocytes, mature T cells, and subsets of activated B cells. It is involved in T-cell regulation and proliferation. Found on 95% of thymocytes and 72% of peripheral lymphocytes, CD5 is also expressed in various T-cell leukemias and lymphomas. Dysregulation is associated with thymus cancer and Richter’s Syndrome.
