Product Specific Information

The 145-2C11 monoclonal antibody reacts with mouse CD3e, a 20 kDa subunit of the TCR complex. The epsilon chain, along with gamma and delta, is essential for proper assembly, trafficking, and surface expression of the TCR complex. CD3 is expressed by thymocytes and all mature T cells.

Binding of 145-2C11 to TCR initiates intracellular signaling leading to activation, proliferation, or apoptosis depending on experimental conditions. Commonly used as a phenotypic marker for mouse T cells.

Each product contains:

  • 1 vial of NovaFluor conjugate
  • 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Mouse splenocytes (≤1.0 µg/test, 100 µL final volume, 10⁵–10⁸ cells/test)

Note:

  • Carefully titrate antibody for optimal performance.
  • NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
  • Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01):

  • Required for optimal staining with NovaFluor conjugates.
  • Add 5 µL per sample before combining with cells.
  • For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology, engineered nucleic acid dye structures for optimized fluorescent signatures minimizing spillover and spread.

Note on Mouse Strains:
NovaFluor Yellow 730 may non-specifically stain B cells in SJL mice. No such staining observed in BALB/c or C57BL/6 mice.


Target Information

The CD3 subunit complex transduces antigen-recognition signals and regulates TCR surface expression. It includes CD3 gamma, delta, epsilon, and zeta subunits, all part of the immunoglobulin superfamily on chromosome 11.

Functions:

  • Mediates T-cell activation and signal transduction
  • Involved in TCR-induced growth arrest, survival, and proliferation
  • Expressed on 68–82% of peripheral blood lymphocytes and 65–85% of thymocytes
  • Not expressed on B or NK cells
  • CD3 gene defects are associated with CD3 immunodeficiency
