Product Specific Information

The 53-6.7 monoclonal antibody reacts with the mouse CD8a molecule, a 32–34 kDa cell surface receptor expressed as CD8αβ heterodimer or CD8αα homodimer.
CD8 binds to MHC class I and associates with protein tyrosine kinase p56lck, playing an essential role in T cell development and activation.

Applications Reported: Flow cytometric analysis.
Applications Tested: Mouse splenocytes; ≤0.5 µg/test (100 µL final volume). Adjust cell number empirically (10⁵–10⁸ cells/test). Titrate for optimal performance.

NovaFluor Dyes Compatibility:

• Not compatible with DNA intercalating viability dyes (PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

Staining Recommendation:
Each NovaFluor conjugate ships with CellBlox Blocking Buffer.
Add 5 µL per sample or per 100 µL of cell suspension (10³–10⁸ cells). Use with antibody cocktails and single-color controls.

Target Information

CD8 (Cluster of Differentiation 8) is a type I transmembrane glycoprotein involved in T cell activation and differentiation.
Expressed predominantly on T cells as CD8αβ heterodimers, it acts as a co-receptor for MHC class I molecules.
CD8α also forms homodimers on subsets of T cells, NK cells, monocytes, and dendritic cells.
Engagement with MHC-I/peptide complexes recruits Lck kinase, leading to CTL activation and pathogen/tumor cell clearance.
Differentiation of naive CD8+ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
