Product Specific Information
The 53-6.7 monoclonal antibody reacts with mouse CD8a, a 32–34 kDa cell surface receptor expressed as either a heterodimer (CD8αβ) or homodimer (CD8αα). CD8 binds to MHC class I and associates with protein tyrosine kinase p56lck, playing a role in T cell development and activation.

- Applications Tested: Flow cytometric analysis of mouse splenocytes (≤0.5 µg/test).
A test corresponds to staining a cell sample in 100 µL volume; cell number may range from 10⁵–10⁸ cells/test.
Antibody titration is recommended for optimal performance.
Compatibility Note:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.
Blocking Recommendation:
Each NovaFluor conjugate ships with CellBlox Blocking Buffer.
Add 5 µL per sample when staining with NovaFluor conjugates, including single-color compensation controls.

Target Information
CD8 (Cluster of Differentiation 8) is a type I transmembrane glycoprotein crucial for T cell signal transduction, differentiation, and activation. It is predominantly expressed on T cells as a CD8αβ heterodimer functioning as a co-receptor for MHC class I molecules. CD8 also exists as CD8αα homodimers on subsets of T cells, NK cells, monocytes, and dendritic cells.
Binding of MHC-I/peptide complexes triggers Lck recruitment, leading to lymphokine production and cytotoxic T lymphocyte (CTL) activation. CTLs are essential for pathogen and tumor cell clearance. Differentiation of naive CD8+ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
