PA1-058 detects prohormone convertase 2 (PC2) from human and mouse alpha pancreas cells.
It has been successfully used in ICC/IF and Western blot procedures.
By Western blot, this antibody detects an ~65 kDa protein representing PC2 from alpha TC 6 cell extract.
This antibody is not recommended for Neuro-2a cells in immunofluorescent applications.

The immunogen is a synthetic peptide corresponding to residues E(622) L D E A V E R S L Q S I L R K N(638) of mouse PC2.
This sequence is completely conserved between mouse and rat protein.
PA1-058 synthetic peptide (PEP-138) is available for use in neutralization and control experiments.

Target Information

The subtilisin-like prohormone convertase (PC) family consists of cellular enzymes that cleave most prohormones and neuropeptide precursors.
They also target numerous other cellular proteins, viral proteins, and bacterial toxins transported by the constitutive secretory pathway for maturation.

PC family members share structural similarities, including:

• A heterogeneous ~10 kDa amino-terminal proregion
• A highly conserved ~55 kDa subtilisin-like catalytic domain
• A heterogeneous carboxyl-terminal domain in length and sequence

These enzymes become catalytically active following proregion cleavage within the appropriate cellular compartment.
Immunolocalization studies show PC1 is found in the perinuclear region and trans-Golgi network, while PC2 is found in the trans-Golgi network and diffusely distributed in the peripheral cytoplasm.
