Product Specific Information

The monoclonal antibody HFR1 recognizes human and mouse epidermal growth factor receptor 4 (ErbB4/HER4).
ErbB4 is part of the receptor tyrosine kinase (RTK) family, which includes HER1 (EGFR), HER2 (ErbB2), and HER3 (ErbB3).
These receptors regulate cell proliferation, differentiation, migration, survival, and apoptosis, and are often dysregulated in cancer.

ErbB4 contains extracellular ligand-binding domains, a transmembrane region, and a cytoplasmic tail with a tyrosine kinase domain and the HFR1 binding site.
Upon ligand binding, ErbB4 can homo- or hetero-dimerize, become phosphorylated, and translocate to the nucleus.
The HFR1 antibody recognizes membrane-bound, cytoplasmic, and nuclear ErbB4.

Cytosolic ErbB4 expression correlates with positive prognosis in certain breast cancer patients, while nuclear ErbB4 expression is inversely correlated with tumor grade and aggressiveness.

Applications Reported: Immunoprecipitation, Western blotting, immunohistochemical staining of formalin-fixed paraffin-embedded tissue, and immunocytochemistry.
Applications Tested: Tested by IHC on formalin-fixed paraffin-embedded human tissue using low pH antigen retrieval at ≤20 µg/mL.
Antibody titration is recommended for optimal performance.


Target Information

Oncogene ERBB4 (HER4) is a type I receptor tyrosine kinase, part of the family including EGFR, ERBB2, and ERBB3.
It binds and is activated by neuregulins and other factors, inducing cellular responses such as mitogenesis and differentiation.

ErbB4 is activated by neuregulins and NTAK, serving as a receptor for heregulin and mediating HGL-stimulated tyrosine phosphorylation.
It regulates metabolism, transcription, cell cycle, cytoskeletal arrangement, movement, apoptosis, and differentiation.

ErbB4 is predominantly expressed in several breast carcinoma cell lines and in normal skeletal muscle, heart, pituitary, brain, and cerebellum.
High mRNA levels are found in T47-D, MDA-MB-453, BT-474, and H3396; intermediate levels in MCF-7, MDAMB-330, and MDA-MB-361; low or absent in MDA-MB-231, MDA-MB-157, MDA-MB-468, and SKBR-3.
Elevated ErbB4 levels in certain tumor cell lines suggest a potential role in malignancy.
