The monoclonal antibody HE12 recognizes human Cyclin E. Cyclins regulate the cell cycle by binding and activating cyclin-dependent kinases (CDK). Cyclin E controls the transition from G1 to S phase by activating CDK2, which phosphorylates downstream proteins facilitating S phase entry. Overexpression of Cyclin E is observed in multiple cancers including breast, colon, bladder, lung, and skin. In breast cancer, its overexpression correlates with higher stage, grade, and poor prognosis.
Applications Reported:
HE12 antibody is used for immunohistochemical staining of formalin-fixed paraffin-embedded tissue sections, microscopy, and immunocytochemistry.
Applications Tested:
Tested in IHC of FFPE human tissue using low pH antigen retrieval; optimal concentration ≤10 µg/mL. Titration recommended for best assay performance.
Target Information
Cyclin E is part of the conserved cyclin family, regulating CDK kinases and controlling the G1/S cell cycle transition. It forms a complex with CDK2, accumulates at the G1-S boundary, and is degraded during S phase. Overexpression contributes to chromosomal instability and tumorigenesis. Cyclin E also associates with NPAT, involved in histone gene expression and cell-cycle progression independent of pRB. Multiple splice variants exist, with two well-characterized transcripts and others not fully sequenced.
