The monoclonal antibody 22C11 recognizes human, mouse, and rat APP (Amyloid Precursor Protein). APP is highly expressed in the central nervous system and exists as three major isoforms due to alternative splicing. It plays roles in synaptic formation, neuronal transport, iron export, and hormonal regulation. Secreted APP (sAPP) may have neuroprotective effects against oxidative stress and excitotoxicity.
APP belongs to a family that includes amyloid precursor-like proteins 1 and 2 (APLP1, APLP2). APLP, particularly APLP2, may share and compensate for APP function. Proteolytic cleavage of APP generates beta amyloid, the main component of senile plaques, a hallmark of Alzheimer’s disease. Abnormal APP processing is also associated with Down’s syndrome, familial Alzheimer’s disease, cerebral hemorrhage, and arthritis.
This 22C11 antibody recognizes pre-A4 and all three isoforms of APP (immature, sAPP, mature) and cross-reacts with APLP2.
Applications Reported: Microscopy and immunocytochemistry.
Applications Tested: Tested by immunocytochemistry of methanol-fixed cells; recommended ≤1 µg/mL. Optimize dilution for specific assay conditions.
Filter Recommendation: For eFluor™ 570 conjugate, use a filter capturing 570 nm emission (e.g., Excitation 545/25, 565LP, Emission 605/70). Alexa Fluor™ 555 or TRITC filters are also suitable.

Target Information
Amyloid Precursor Protein (APP) functions as a cell surface kinesin I membrane receptor mediating axonal transport of beta-secretase and presenilin 1. It is essential for neurite growth, neuronal adhesion, and axonogenesis. APP (100–140 kDa) exists as several isoforms from alternative splicing. Proteolytic cleavage by beta- and gamma-secretases generates beta amyloid, the primary component of senile plaques in Alzheimer’s disease. Abnormal APP processing is also linked to Down’s syndrome, early-onset familial Alzheimer’s disease, and cerebral hemorrhage.
