Product Specific Information

The monoclonal antibody 1A4 recognizes human, mouse, and rat alpha-smooth muscle actin (α-SMA), a 42 kDa protein that forms part of the cytoskeletal network. This antibody is specific for the alpha isoform expressed by smooth muscle cells, myofibroblasts, and myoepithelial cells. It is useful for identifying leiomyomas, leiomyosarcomas, pleomorphic adenomas, and serves as a prognostic marker for basal cell carcinoma.

Applications Reported:
Used in immunohistochemistry (frozen and paraffin-embedded tissue), microscopy, and immunocytochemistry.

Applications Tested:
Validated for IHC of formalin-fixed paraffin-embedded human tissue using low pH antigen retrieval, effective at ≤1 µg/mL. Optimal concentration should be determined per assay.

Filter Recommendation:
For eFluor™ 570 conjugate, use filters capturing 570 nm emission (e.g., Excitation 545/25, 565LP, Emission 605/70). Compatible with Alexa Fluor™ 555 or TRITC filters.

Target Information

Smooth Muscle Actin (α-SMA) is part of the actin protein family involved in cell motility, structure, and integrity.

• Alpha actin: major component of contractile apparatus
• Beta and gamma actins: regulate cell motility

Smooth muscle actin is found in skeletal muscle, vessel walls, gut wall, myometrium, and myoepithelial cells. It interacts with myosin for muscle contraction and cell movement.
Defects in the gene can cause familial thoracic aortic aneurysm type 6.
Alpha-smooth muscle actin is abundant in vascular and visceral smooth muscle, and also appears in fibroblastic stress fibers during wound healing and fibrocontractive diseases.
