Target Information

MYLK (MLCK) is a serine/threonine kinase that phosphorylates myosin regulatory light chains to assist in contractile activity (myosin-actin interaction).
MLCK mediates smooth muscle contraction via phosphorylation of a specific serine residue in the N-terminus of myosin light chains (MLC), facilitating myosin interaction with actin filaments.
It plays a key role in vascular permeability, tissue edema formation, and in the nervous system contributes to astrocytic process growth and neurotransmitter release.
MLCK also participates in fibroblast apoptosis signaling.
Smooth muscle and non-muscle isozymes are expressed broadly across adult and fetal tissues.
Non-muscle isoform 2 is the dominant splice variant, and the Telokin isoform binds calmodulin.
MLCK is likely down-regulated by phosphorylation and contains one fibronectin type III domain and nine immunoglobulin-like C2-type domains.
