Target Information
Glutamate is the main excitatory neurotransmitter in the brain. It was long thought to act only on ligand-gated receptor channels such as NMDA, AMPA, and kainate receptors, which mediate fast excitatory synaptic transmission.
Recent findings show that glutamate also regulates enzymes producing second messengers via specific G-protein-coupled receptors, known as metabotropic glutamate receptors (mGluRs).
Group-I mGluRs (including mGluR1a and mGluR5) stimulate phospholipase C, leading to increased phosphoinositide turnover and calcium release from internal stores.
Group-II and Group-III receptors (including mGluR2 and mGluR3) are coupled to inhibition of adenylyl cyclase.
